Hmm, there's a lot of schools of thought to this. You could go with Colour of Magic and read in publication order but the earlier few aren't his best (obviously; he was just starting out).

My tip would be you start at Guards Guards, that's an all-round classic that doesn't need much introduction. It sets out the flow of his books, his style of humour, and it kicks off one of the mini-series that focusses on the City Watch if you want to just continue reading a set of characters.
