Yes I totally agree that the authors did not resolve the discrepancies, of which I see two. The reference sample is supposed to be 70-80kDa and it’s >100 kDa. The sample also has two prominent bands one of which could plausibly be the correct mass given the low resolution of the gel and the shift in the recombinant control. Given that the detection antibody identifies both bands it is likely the smaller one is a truncated version of the other. Most likely a degradation product? That being said you’d need to do ALOT more work to show that.
The other issue is separate from this and I agree that reproducibility from lot to lot is paramount.
