I think it's mostly straight forward. It's basically Bill Pullman suspecting his wife is having an affair, killing her and then imagining himself living the life of a younger, sexually potent young man but ultimately getting lost in his fantasy. Depending on how you interpret that final scene, it's either his last moments before he gets the electric chair or its a loop that restarts. Most people usually believe the former and I would, too.
The one element I admittedly don't fully understand is the "Dick Laurent is dead" scene in the beginning, because it's hard to know if he really imagined that or partly imagined that or if Dick Laurent is even real. My interpretation of that scene was that it was a meta moment/fourth wall break from which the rest of the story commences. Like from the moment that ends the real Fred begins imagining that whole sequences of events and the Dick Laurent is dead opening occasionally bleeds into it (as seen in the scene after he talks to the mystery man and when he goes back to his house at the end.)

I actually find Mulholland Drive to be easier to understand than Lost Highway, but then the mystery of Dick Laurent is dead and the ending is what captives me all these years later.
